Buster Busts Loose is a side-scrolling platformer for the Super Nintendo starring Buster Bunny. The stages are framed as a series of cartoon shoots, moving through settings such as the Acme Looniversity school grounds, a western, a sci-fi episode and a football field, each with its own hazards and enemies. Buster's main attacks are a jump kick and a dash that builds up a limited run meter, letting him charge through enemies, run up walls and cover long gaps when timed correctly. Other Tiny Toons characters turn up as helpers, bosses and cameo appearances between stages. Bonus mini-games can be played after clearing sections, and the game uses large, expressive sprites and animation to imitate the look of the television series.
